Caenorhabditis elegans

Caenorhabditis elegans is an experimental nematode model organism used to probe conserved mechanisms of aging, stress resistance, neurobiology, and toxicology. In this system, ethanol exposure affects sensorimotor behavior through dopaminergic dop 3 and serotonergic tph 1 pathways, while mitochondrial longevity is studied using long-lived clk 1 and isp 1 mutants under persistent mitochondrial inefficiency. It is also widely used to test natural products and small molecules for lifespan and healthspan effects, including bisdemethoxycurcumin, corylin, psp 1 1, geopropolis, eurycoma longifolia jack polysaccharides, iridoid glycosides, and dietary phosphatidylcholine. Recent studies report that corylin activates a DAF-16/SKN-1-coordinated autophagy–mitochondrial homeostasis axis, and that phosphatidylcholine synthesis decline is a malleable trigger of natural mitochondrial aging. Overall, C. elegans serves as a fast in vivo platform for aging biology, oxidative stress resistance, neuroprotective screening, and mitochondrial stress-response research.

Aging and longevity

  • Corylin promoted longevity in C. elegans through coordinated activation of the DAF-16 and SKN-1 pathways and an autophagy–mitochondrial homeostasis axis. (PMID:41730400)
  • Bisdemethoxycurcumin extended lifespan and healthspan via modulation of EGFR-linked signaling pathways. (PMID:41804763)
  • Terbinafine and miglustat extended lifespan and improved healthspan in the nematode model. (PMID:41913053)
  • Eurycoma longifolia Jack polysaccharides showed aging-modulatory effects in C. elegans, supporting its use as a longevity screen. (PMID:41954810)

Stress resistance and healthspan

  • Two newly described iridoid glycosides improved healthspan, with stress endurance mediating the effect. (PMID:41690035)
  • Geopropolis extracts enhanced oxidative stress resistance and increased lifespan in C. elegans. (PMID:41901279)
  • PSP-1-1 from steamed Polygonatum sibiricum showed anti-aging and neuroprotective effects in the worm model. (PMID:41943329)
  • Dietary phosphatidylcholine boosting restored late-life mitochondrial integrity in vivo, linking lipid homeostasis to aging resilience. (PMID:42000749)

Neurobiology and toxicology

  • Ethanol exposure was used to assess sensorimotor effects in C. elegans, with dopaminergic dop 3 and serotonergic tph 1 genes contributing to the response. (PMID:41871151)
  • PSP-1-1 was evaluated for neuroprotective activity alongside its anti-aging effects. (PMID:41943329)
  • Geopropolis was tested not only for lifespan effects but also for resistance to oxidative stress, a common toxicological readout. (PMID:41901279)
  • C. elegans remains a preclinical model for screening compounds that alter behavior, stress tolerance, and organismal survival. (PMID:41871151)
